Assay of Paracetamol-500mg and 650mg- By UV Spectrophotometric method

1.0 OBJECTIVE

To provide the procedure for Assay of Paracetamol-500mg and 650mg- By UV Spectrophotometric method

2.0 SCOPE

This procedure is Applicable to Assay of Paracetamol-500mg and 650mg- By UV Spectrophotometric method

3.0 RESPONSIBILITY

3.1 QC-Chemist

4.0 ACCOUNTABILITY

4.1 Executive – Quality control

4.2 Head – Quality control

5.0 PROCEDURE

5.1 Each Uncoated tablet contains:

paracetamol    IP    500 mg/650 mg

5.2 Assay

5.2.1 Standard Solution :

Weigh accurately 75mg of Paracetamol working standard in to a 100ml volumetric flask, dissolve in 50ml of 0.1M Hcl sonicate for 3minutes and makeup the remaining volume with 0.1M Hcl. From the resulting solution take 5ml and dilute to 50ml with 0.1M Hcl shake well and from this solution again take 5ml and dilute to 50ml with 0.1M Hcl.

5.2.2 Sample Solution:

Weigh accurately tablet powder equivalent to 75mg of Paracetamol in to a 100ml volumetric flask, dissolve in 50ml of 0.1M Hcl sonicate for 3minutes and makeup the remaining volume with 0.1M Hcl. Filter the sample Solution with whatman filter paper and  reject first few ml of the filtrate and from the filtrate take 5ml and dilute to 50ml with 0.1M Hcl shake well and from this resulting solution again take 5ml and dilute to 50ml with 0.1M Hcl.

5.2.3 Measure the absorbance of the resulting solution at the maximum at 244nm using 0.1M Hcl as blank. Calculate the amount of Paracetamol by comparing the sample values with standard values.

Sample absorbance    x  Standard weight  x Assay x Average weight of the tablet =
Standard absorbance  x  Sample Weight   x   100

= ____________x100
