Micropipette operation and calibration procedure

    1.0 OBJECTIVETo describe a procedure for Calibration of Micropipette by using water.

    2.0 SCOPE

    This procedure is applicable to Calibration of Micropipette and is applicable in Microbiology

    3.0 RESPONSIBILITY

    3.1 Microbiologist /Officer is responsible for the execution. 

    3.2 HOD or designee is responsible for review, effective implementation and compliance to the calibration of micropipette for execution. 

    4.0 PROCEDURE

     



    4.1 Operation

    4.1.1 Set the required volume by rotating color knob clockwise and anti clockwise.

    4.1.2 Fix the micro tip by applying light pressure.

    4.1.3 Press the Plunger and insert the micro tip in the solution to be taken.

    4.1.4 Release the plunger to get the required solution.

    4.1.5 Transfer the solution by pressing the plunger little more than the normal. Press the dispenser knob to remove the used micro tip.

    Note: Use this pipette (100 -1000µl) for volume not less than 100 µl and     not more than 1000 µl Use this pipette (1 -10ml) for volume not less than 1ml and not more than 10ml.

    4.2 Calibration

    4.2.1 Set the volume at 200 µl, 500 µl and 1000µl for micropipette (T -1000µl).

    4.2.2 Follow the operation procedure from point no 6.1.2 to 6.1.5.

    4.2.3 Take the water at room temperature as per set volume and weigh it in a weighing bottle, which is previously dried and weighed.

    4.2.4 Record the reading.

    4.2.5 Repeat the analysis for 5 times and take the average.

    4.2.6Calculate the volume by dividing the average weight of water with the actual volume from the table given as per the test temperature and record the values in Attachment #1

    4.2.8 Precautions :

    4.2.8.1 After fixing volume of micropipette don’t rotate the push button.

    4.2.8.2  No foreign particle should be present in the tip as this can block the delivery of the liquid and this can result in incorrect volume dispenses.
